چکیده

The RNA 3 of rice stripe virus (RSV) isolate T was cloned and its nucleotide sequence was determined. The complete primary structure was found to consist of 2504 nucleotides. One putative open reading frame (ORF), between nucleotides 2412 and 1444 in the 5'-proximal region of the virus complementary-sense RNA, encoded a 322 amino acid protein with an Mr of 35,134 that was identified as the coat protein. The other ORF, between nucleotides 66 and 701 in the 5'-proximal region of the virus-sense RNA, encoded a 211 amino acid protein with an Mr of 23,874. An intergenic non-coding region (742 nucleotides) between the two ORFs contains oligo(A)- and oligo(U)-rich regions that can be arranged into a 126 base pair stem configuration. These results suggest that RSV RNA 3 has an ambisense coding strategy.

Ambisense segment 3 of rice stripe virus: the first instance of a virus containing two ambisense segments.

The complete nucleotide sequence of rice stripe virus (RSV) segment 3 shows that it has two open reading frames, one in the viral-complementary sequence, which codes for the nucleocapsid protein, and the other in the viral-sense sequence. The non-coding region between the ambisense genes in RSV segment 3 contains several U and A tracts, as do the ambisense S segments of phleboviruses and uukuvi...

In vitro Expression Studies of Three Proteins of Iranian Wheat Stripe Virus

The genome of Iranian wheat stripe virus (IWSV), a tentative member of the genus Tenuivirus, is comprised of three ambisense and one negative sense RNA segments. The coat and non-structural proteins encoded by the vcRNA3 and vRNA4 genes, respectively, were efficiently translated in vitro. Translated proteins of vcRNA3 and vRNA4 transcripts were approximately 35000 and 22000 in Mr, respectively,...

Ambisense segment 4 of rice stripe virus: possible evolutionary relationship with phleboviruses and uukuviruses (Bunyaviridae).

The complete nucleotide sequence of segment 4 of the rice stripe virus (RSV) genome was determined from overlapping cDNA clones and by direct RNA sequencing. The segment has two long open reading frames (ORFs). One of the ORFs (534 bases) is in the 5' region of the viral sequence, and the other (858 bases) is in the viral complementary sequence of the viral 3' region. Such ambisense genome orga...

Complete sequence of maize stripe virus RNA4 and mapping of its subgenomic RNAs.

The complete nucleotide sequence of maize stripe virus RNA4 was determined and found to consist of 2227 nucleotides containing two significant open reading frames. One, in the 5' end of the viral RNA, encodes the major non-capsid protein of M(r) 19,815. The other is located in the 5' end of the viral complementary RNA and could encode a protein of M(r) 31,900. This protein has not been identifi...

Nucleotide sequence of RNA 1, the largest genomic segment of rice stripe virus, the prototype of the tenuiviruses.

The complete nucleotide sequence of RNA 1, the largest genomic segment of rice stripe virus (RSV), was determined using two sets of overlapping cDNA clones. RNA segment 1 comprises 8970 nucleotides and on the viral complementary sequence has a single long open reading frame coding for a protein of 2919 amino acids with an estimated M(r) of 336860. Amino acid sequence comparisons of the putative...
